ANTIOXIDANT ACTIVITY TEST of ETHER FRACTION ETHANOL EKSTRACT FROM A GINGER ROOT (Languas galanga L. Stuntz) USING DPPH (1,1-difenil-2-pikrilhidrazil) METHODS

    The science and technology development and also the change of community lifestyle cause some various appearances of degenerative diseases. Degenerative diseases possible vaguely if body have a free radical scavenger such as antioxidant.
A Ginger root (Languas galanga L. Stuntz) contains flavonoid that have an antioxidant activity. This research was performed to recognize antioxidant activity of ether fraction ethanol extract from a Ginger root using DPPH ( 1,1-defenil- 2- pikrilhidrazil) methods.

    Ether fraction ethanol extract of a Ginger root (as a sample) was obtained by Soxhlet instrument with petroleum ether solution to eliminate the non polar compound, followed by Soxhlet instr ument with ethanol 96% the later with ether fraction. Antioxidant activity using DPPH method was performed by spectrophotometer with maximum wavelength of 513,5 nm. The concentration of samples uses were 20 µg/ml; 40 µg/ml; 80 µg/ml; 160 µg/ml and the positive control quersetin 1 µg/ml; 2 µg/ml; 4 µg/ml; 8 µg/ml; 16 µg/ml. Percentage of free radical scavenging and EC50 value were determined.The research showed that all treatment has an antioxidant activity as a free radical scavenger. Percentage of free radical scavenging was in a sample of concentration 20µg/ml ( 13,41+ 4,55)%; 40 µg/ml (25,84 + 3.02)%; 80 µg/ml ( 44.20 + 2,51)%, and 160 µg/ml ( 65,95 + 1,30)%. Whereas the positive control quersetin was in a concentration 1 µg/ml (12,23 + 8,68)%; 2 µg ( 17,03 + 5,75)%; 4 µg/ml ( 25,38 + 5,63)%; 8 µg ( 52,22 + 5,86)%; and 16 µg ( 80,15 + 1,53)%. EC50 value for sample amount 109,73 µg/ml and for positive control quersetin amount 8,92 µg/ml. From test of statistic one way Anava with 95% of significance which continued by Tukey test andLSD, showed that there were significant differences between all treatment. On the concentration sample 20 µg/ml and concentration quersetin 1 µg/ml and concentration sample 40 µg/ml and concentration quersetin 4 µg/ml show that there were no significance difference.
