TEST ethanol extract antifungal activity

    Antifungal agent is compound which is used for infection treatment which caused by fungi. Nigella sativa L. seed has many advantages and in the traditionally therapy can be used to cure disease what has been caused by microbial. Based on case, the aim of this research to know the antifungal activity of Nigella sativa L. seed ethanol extract against Trichophyton rubrum and to identify the chemical contain alkaloid, saponin and volatile oil.

    The determination MIC (Minimum Inhibitory Concentration) conducted by using liquid method, that is with ethanolic extract solution which have been added fungi suspension 106 CFU/ml, to determine the MFC (Minimum Fungisid Concentration) conducted by inscribing result incubated at SDA media,. Ethanolic extract concentration used for Trichophyton rubrum in this research were 7,5%; 6,25%; 5,0%; 3,75%; 2,5% w/v. Identification of chemical compound was carried out by thin layer Chromatography with methanol:kloroform (9:1) as mobile phase for alkaloid and saponin; benzene as mobile phase for volatile oil. The UV366 nm light, UV254 nm light, Vanillin sulfat acid and Dragendorff, were used to detect the Chromatography profile.
    The result showed that the value of Minimum Inhibitory Concentration (MIC) could not be known and turbid while the Minimum Fungisid Concentration (MFC) of Nigella sativa L. seed ethanolic extract against Trichophyton rubrumnwas 5,0% w/v. The result thin layer chromatography showed that Nigella sativa L.seed ethanolic extract contained alkaloid, saponin and volatile oil
