The damages in cells and in tissues are the inhibitors of most of diseases. Such damages are caused by free radical. Anticipation toward the formation of free radical in human body was conducted by consuming the antioxidant sources. One of natural compounds that is famous because it has activity as antioxidant is flavonoid. The main purpose of this research is to compare free radical scavenger activity of ether and water fraction of ethanol extract of Centella asiatica herbs.
The dry powder of Centella asiatica herbs was extracted by 70% of ethanol by maceration method. The thick extract that gained was treated fractionation with ether water. Fraction of water and fraction of ether then were examined on its activity of antioxidant quantitatively that was determined by DPPH method (1, 1-difenil-2-pikrilhidrazil). The series of contents used in this research were 40; 60; 80; 100; 120 µg/ml to ether fraction ethanol extract of Centella asiatica herbs, 20; 40; 60; 80; 100 µg/ml to water fraction ethanol extract of Centella asiatica herbs, and 3; 4; 5; 6; 7 µg/ml to quersetin as a positive control. Data was gained then to be analyzed by using statistical test by two way ANOVA with 95% of confident level. The activity of free radical scavenging DPPH in ether and water fraction of ethanol extract of Centella asiatica herbs, and also quersetin could be stated by ES50 parameter.
The percent result of free radical scavenger activity of water fraction ethanol extract of Centella asiatica herbs in each concentration were 20 µg/ml (16,56±0,69), 40 µg/ml (35,22±0,37), 60 µg/ml (46,86±0,25), 80 µg/ml (56,33±0,46), 100 µg/ml (57,74±0,26). Eter fraction ethanol extract of Centella asiatica herbs were 40 µg/ml (14,79±0,45), 60 µg/ml (22,33±0,13), 80 µg/ml (31,10±0,53), 100 µg/ml (37,06±0,20), 120 µg/ml (46,53±0,15). In quersetin 3 µg/ml (24,15±0,91), 4 µg/ml (31,54±0,65), 5 µg/ml (36,68±1,25), 6 µg/ml (43,80±1,83), 7 µg/ml (56,12±0,36), the result be found of differences between each treatment. The value of ES50 quersetin 6,46 µl/ml, the water fraction of ethanol extract of Centella asiatica herbs 74,42 µg/ml, and the ether fraction of ethanol extract of Centella asiatica herbs 130,22 µg/ml.
